Pharmasave Katherine Pharmacy in Oasis Shopping Centre Manager Rebecca Paine said they were "fully booked out every day for appointments", especially since vaccination mandates came into effect.
The pharmacy began offering COVID-19 vaccinations in late September.
"It was more 'You have to be vaccinated to keep your job. That's when our bookings started filling up."
Ms Paine said the pharmacy, which is one of two in town, has been flat out during the lockdown as a number of staff members were in isolation due to being close contacts of COVID cases.
More Katherine lockdown news:
"We do need all our staff. I mean, at any time, things can change," she said.
Some of our staff have been close contacts."
She said a significant problem with the lockdown was people from remote cattle stations in the Big Rivers region being able to get medication and supplies.
"It's the people from stations three, four hours away who can't actually get here," Ms Paine said.
"There's not a lot we can do for them. So they've got to get people to come here for them and meet them somewhere. That's been a bit tricky."
